Abstract

Triphala a Ayurvedic preparation, is taken as a general health tonic and has potent antioxidant with anticancer properties. Triphala churna is effective in high doses and the present study is undertaken to prepare nanoformulation of triphala extract and to study its effectiveness in low doses. Nanoformulation of methanolic extract of triphala churna (TCNF) was prepared by modified solvent evaporation technique using 1:3 drug polymer ratio. Polyvinylalcohol (PVA) was used as a polymer as well as stabilizer. The mean particle size and zeta potential was found to be 317 nm and -23.8 mV respectively. FTIR and DSC analysis revealed the absence of any interaction between triphala extract and excipients during formulation development process. HPTLC confirmed the presence of gallic acid and ensured the chemical stability of the nanoparticles of triphala. Nanoformulation of triphala was evaluated for its in vitro antioxidant activity by ABTS, DPPH and nitric oxide scavenging assays. The nanoformulation of triphala has shown the reduction in IC50 values in all the tested models. SRB assay was performed by in vitro method in which nanoformulation of triphala has shown better cytotoxic activity. These findings provide further understanding for the increase in therapeutic efficacy of nanoformulation of triphala.
