Abstract
A mixture of ct-N-benzoyl-L-arginine and etbyl-ex-N-benzoyl-L-argininate could be separated on a 5 ml column of Sephadex G-15. While benzoyl arginine was eluted witb water, tbe ester was bound to tbe column and could be eluted with 0.05 M NaCl. This differential behaviour was used in the development of a colorimetric assay method for trypsin. The enzyme action on ethylex-N-benzoyl-L-argininate at pH 7.6 was stopped by the addition of trichloroacetic acid, and an aliquot of the supernatant was passed through a column of Sephadex G-15. ct-N-Benzoyl-L-arginine was eluted with water and assayed by the Sakaguchi method.
