Abstract

The purpose of this research work was to formulate an anti diabetic and anti hypertensive in a single dosage form. Metformin HCl in sustained release layer and Ramipril in immediate release layer of the bilayer tablet.The tablets were prepared using Hydroxypropylmethylcellulose (HPMC K4M & HPMC K100M) and Polyethylene oxide (Polyox N-70, Polyox N-75, Polyox WSR-301) as release retarding polymers in various combination and concentrations. The effect of different super disintegrants on immediate release in various concentrations was also studied. The Drug Excipient compatibility studies like FTIR spectroscopy and Differential scanning calorimetry showed the absence of drug-drug and drug excipients interaction. Tablets were prepared by Direct Compression technique and found within the acceptable limits for various physiochemical parameters such as thickness, hardness, friability, weight variation, drug content, in vitro dissolution studies and stability studies. In vitro release studies were carried out using the dissolution apparatus II at 100 rpm in Phosphate buffer pH 6.8 for Metformin HCl and at 75 rpm in 0.1N HCl for Ramipril as described in USP 2000. The analytical methods used were U.V spectroscopy and HPLC for Metformin HCl and Ramipril respectively. The optimized formulation was found to be stable at accelerated storage conditions for 1 month with respect to appearance. The drug content of Metformin HCl in the formulation was well within the limits but Ramipril showed degradation and it was found to the same extent as marketed tablet.
